A comparison of behavioral and reproductive parameters between wild-type, transgenic and mutant zebrafish: Could they all be considered the same “zebrafish” for reglementary assays on endocrine disruption?

چکیده

Transgenic zebrafish models are efficiently used to study the effects of endocrine disrupting chemicals (EDC); thereby informing on their mechanisms action. However, given reported differences between strains at genetical, physiological and behavioral levels; care should be taken before using these transgenic for EDC testing. In present study, we undertook a set experiments in different and/or mutant interest testing: casper, cyp19a1a-eGFP, cyp19a1a-eGFP-casper, cyp11c1-eGFP, cyp11c1-eGFP-casper. Some traits, some biochemical reproductive endpoints commonly testing were assessed compared those obtained WT AB ensure that transgene insertion mutations do not negatively modify basal physiology or behavior fish. Behavioral traits considered as anxiety sociality have been monitored. Sociality was evaluated by monitoring time spent near congeners shuttle box while Novel tank diving test. No critical difference observed either level. Concerning reproduction, no significant number eggs laid per female, viability female circulating VTG concentrations noted 5 transgenic/mutants studied. summary, had influence measured conditions. These results prerequisite use transgenic/mutant Next step will determine sensitivity biological chemical exposure accurately validate existing fish assays

Behavioral visual responses of wild-type and hypopigmented zebrafish

Zebrafish possess three classes of chromatophores that include iridophores, melanophores, and xanthophores. Mutations that lack one or two classes of chromatophores have been isolated or genetically constructed. Using a behavioral assay based on visually mediated escape responses, we measured the visual response of fully and partially pigmented zebrafish. In zebrafish that lack iridophores (roy...
